Racional: As aderências induzidas por biomateriais implantados experimentalmente na cavidade abdominal são estudadas basicamente com o reparo primário de diferentes defeitos produzidos na parede abdominal ou com a correção de hérnias incisionais realizadas previamente, sem definição precisa do modelo mais adequado. Objetivo: Descrever as aderências que ocorrem após o desenvolvimento de hérnias incisionais, antes do implante de próteses, em modelo experimental para estudo de alterações induzidas por diferentes biomateriais. Métodos: Foram realizadas hérnias incisionais em 10 ratos, com orifícios herniários de dimensões padronizadas, obtidos pela incisão mediana da parede abdominal e eversão das bordas do defeito. Após 10 dias foram evidenciadas aderências de estruturas abdominais quando as hérnias foram reparadas com diferentes próteses. Resultados: Foi possível evidenciar hérnias com saco herniário bem definido em todos os ratos já no décimo dia após o procedimento inicial. Aderências do omento maior ocorreram em cinco animais, dos quais dois também apresentaram aderências de alças do intestino delgado além do omento, e outros dois aderências do fígado juntamente com o omento maior, números com significância estatística pelo teste t de Student (p<0,05). Conclusão: A opção por modelo experimental de reparo de hérnia incisional previamente induzida, embora mimetize a situação clínica real, implica em aderências importantes, com possível repercussão na avaliação da segunda operação, quando são utilizados diferentes implantes de material sintético.

Background: Adhesions induced by biomaterials experimentally implanted in the abdominal cavity are basically studied by primary repair of different abdominal wall defects or by the correction of incisional hernias previously performed with no precise definition of the most appropriate model. Aim: To describe the adhesions which occur after the development of incisional hernias, before the prosthesis implantation, in an experimental model to study the changes induced by different meshes. Methods: Incisional hernias were performed in 10 rats with hernia orifices of standardized dimensions, obtained by the median incision of the abdominal wall and eversion of the defect edges. Ten days after the procedure adhesions of abdominal structures were found when hernias were repaired with different meshes. Results: The results showed hernia sac well defined in all rats ten days after the initial procedure. Adhesions of the greater omentum occurred in five animals of which two also showed adhesions of small bowel loops besides the omentum, and another two showed liver adhesions as well as the greater omentum, numbers with statistical significance by Student's t test (p<0.05). Conclusion: Although it reproduces the real clinical situation, the choice of experimental model of incisional hernia repair previously induced implies important adhesions, with possible repercussions in the evaluation of the second operation, when different implants of synthetic materials are used.

RACIONAL: A ligadura parcial da veia porta acarreta aumento da pressão portal que se mantém estável mesmo após o aparecimento da circulação colateral, havendo adaptação funcional à diminuição prolongada do fluxo sanguíneo hepático. OBJETIVO: Avaliar se diferentes taxas de constrição produzidas por esta ligadura interferem em modelo experimental no rato. MÉTODOS: Foram utilizados três grupos de cinco animais; no grupo 1 (sham-operated) foi realizada dissecção e medida do diâmetro da veia porta. A hipertensão portal foi induzida pela ligadura parcial da veia porta, reduzindo seu calibre para 0,9 mm nos 10 animais restantes, independente do diâmetro inicial das veias. Cinco animais com hipertensão portal (grupo 2) foram reoperados após 15 dias e os ratos do grupo 3 após 30 dias. O cálculo da taxa de constrição foi realizado através de fórmula matemática específica (1 - π r2 / π R2) x 100% e a análise estatística com o teste t de Student. RESULTADOS: O diâmetro médio inicial da veia porta dos animais foi de 2,06 mm, com taxa de constrição média de 55,88%; embora o diâmetro das veias e a taxa de constrição no grupo 2 tenha sido menor que no grupo 3 (2,06 mm - 55,25% e 2,08 mm - 56,51% respectivamente), em todos conseguiu-se induzir hipertensão portal e não foram encontradas diferenças macroscópicas significativas entre os animais reoperados após 15 ou 30 dias, sendo considerado o prazo menor suficiente para avaliação. Quando comparados o diâmetro inicial da veia e a taxa de constrição realizadas nos grupos 2 e 3 não houve significância estatística (p>0,05). CONCLUSÃO: A hipertensão portal pré-hepática no rato pode ser induzida pela redução de calibre da veia porta para 0,9 mm de diâmetro, independente do diâmetro inicial da veia e da taxa de constrição a que este vaso foi submetido.

BACKGROUND: Partial portal vein ligation causes an increase in portal pressure that remains stable even after the appearance of collateral circulation, with functional adaptation to prolonged decrease in portal blood flow. AIM: To assess whether different constriction rates produced by partial ligation of the vein interfere with the results of this experimental model in rats. METHODS: Three groups of five rats each were used; in group 1 (sham-operated), dissection and measurement of portal vein diameters were performed. Portal hypertension was induced by partial portal vein ligation, reducing its size to 0.9 mm in the remaining 10 animals, regardless of the initial diameter of the veins. Five animals with portal hypertension (group 2) underwent reoperation after 15 days and the rats in group 3 after 30 days. The calculation of the constriction rate was performed using a specific mathematical formula (1 - π r 2 / π R2) x 100% and the statistical analysis with the Student t test. RESULTS: The initial diameter of the animal's portal vein was 2.06 mm, with an average constriction rate of the 55.88%; although the diameter of the veins and the constriction rate in group 2 were lower than in group 3 (2.06 mm - 55,25% and 2.08 mm - 56.51%, respectively), portal hypertension was induced in all rats and no significant macroscopic differences were found between the animals that were reoperated after 15 days and after 30 days respectively, being the shorter period considered enough for the evaluation. Comparing the initial diameter of the vein and the rate of constriction performed in groups 2 and 3, no statistic significance was found (p>0.05). CONCLUSION: Pre-hepatic portal hypertension in rat can be induced by the reduction of the portal vein diameter to 0.9 mm, regardless the initial diameter of the vein and the vessel constriction rate.

RACIONAL: O resultado negativo de uma pesquisa nem sempre indica falha, e quando os dados obtidos não permitem uma conclusão adequada, ou ainda, são contrários ao projeto inicial, não devem simplesmente ser desprezados e arquivados. OBJETIVO: Relatar falha ao realizar em animais de pequeno porte modelo experimental de isquemia e reperfusão hepática normotérmica, contínua ou intermitente, visando estudar parâmetros bioquímicos e histológicos após a recuperação pós-operatória. MÉTODOS: Quinze ratos da linhagem Wistar foram divididos em três grupos com cinco animais cada; todos foram operados, a cavidade abdominal foi suturada após os procedimentos propostos para cada grupo e os ratos foram observados por 6 h ou até morrerem, quando foram reoperados. Foram realizados no Grupo 1, controle (sham-operated): dissecção do hilo hepático; no Grupo 2: pinçamento do hilo hepático por 30 m; no Grupo 3: pinçamento do hilo hepático por 15 m, reperfusão do fígado por 5 m e mais 15 m de pinçamento. Os dados dos grupos 2 e 3 foram comparados com o teste t de Student. RESULTADOS: Todos os animais do Grupo 1 sobreviveram 6 h. Dois do Grupo 2 morreram antes das 6 h necessárias para a validação do experimento, dois não se recuperaram da primeira anestesia e um sobreviveu até o final. No Grupo 3, quatro animais morreram antes das 6 h previstas e um sobreviveu o tempo necessário. Apenas um animal do Grupo 2 e um do Grupo 3 sobreviveu ou se encontrava em condições para a complementação do estudo. Não houve significância estatística quando os resultados dos Grupos 2 e 3 foram comparados (p>0,05). CONCLUSÃO: A morte de seis animais antes do período de observação necessário após a primeira operação inviabilizou a proposta inicial do experimento.

BACKGROUND: The negative result of a research does not always indicate failure, and when the data do not permit a proper conclusion, or are contrary to the initial project, should not simply be discarded and archived. AIM: To report failure after performing experimental model of liver ischemia and reperfusion normothermic, continuous or intermittent, in small animals aiming at the study of biochemical and histological parameters after postoperative recovery. METHODS: Fifteen Wistar rats were divided into three groups of five animals each; all underwent surgery, the abdomen was sutured after the proposed procedures for each group and the animals were observed for 6 h or until they died, and then were reoperated. In Group 1, control (sham-operated): dissection of the hepatic hilum was performed; in Group 2: clamping of the hepatic hilum for 30 m; in Group 3: clamping of the hepatic hilum for 15 m, reperfusion for 5 m and another 15 m of clamping. Data from Groups 2 and 3 were compared with Student's t test. RESULTS: All animals of Group 1 survived for 6 h. Two animals in Group 2 died before the 6 h needed to validate the experiment; two did not recover from anesthesia and one survived until the end. In Group 3, four animals died before the 6 h established and one of them survived the required time. Only one animal in Group 2 and one in Group 3 survived and were able to accomplish the study. There was no statistical significance when the results of Groups 2 and 3 were compared (p>0.05). CONCLUSION: The death of six animals before the necessary period of observation turned the initial proposal of the experiment unfeasible.

RACIONAL: O cálculo da relação de volumes entre o saco herniário e a cavidade abdominal em hérnias incisionais é feito com base em cortes tomográficos e com a fórmula matemática do volume da elipsóide, permitindo determinar se a hérnia é gigante ou se há "perda de domicílio". Como as imagens utilizadas não são figuras geométricas exatas, foi realizado o estudo do volume de dois órgãos sólidos de ratos da linhagem Wistar, para validar estes cálculos. OBJETIVO: Correlacionar dois métodos para determinação do volume do rim e do baço de ratos, comparando um método direto de avaliação do volume com o cálculo matemático deste valor. MÉTODOS: O volume do rim esquerdo, geometricamente mais regular, e do baço, com seu formato peculiar, de dez animais foram estabelecidos em centímetros cúbicos após imersão total em água, com auxílio de proveta graduada em milímetros; estes valores foram comparados com os obtidos através do cálculo do mesmo volume com fórmula matemática específica: V= 4/3 x π x (r1 x r2 x r3). Os dados obtidos foram comparados e submetidos à análise estatística pelo teste t de Student. RESULTADOS: Embora o volume médio obtido tenha sido maior com o método direto, 1,13 cm3 para o rim esquerdo e 0,71 cm3 para o baço, do que os valores calculados com a fórmula matemática, 0,81 cm3 e 0,54 cm3 respectivamente, não houve significância estatística entre as diferenças dos valores encontrados para os dois órgãos (p>0,05). CONCLUSÃO: Houve correlação adequada entre o cálculo direto do volume do rim e do baço com o resultado do cálculo matemático destes valores nos animais estudados.

BACKGROUND: The calculation of the volume ratio between the hernia sac and the abdominal cavity of incisional hernias is based on tomographic sections as well as the mathematical formula of the volume of the ellipsoid, which allows determining whether this is a giant hernia or there is a "loss of domain". As the images used are not exact geometric figures, the study of the volume of two solid organs of Wistar rats was performed to validate these calculations. AIM: To correlate two methods for determining the volume of the kidney and spleen of rats, comparing a direct method of observation of the volume with the mathematical calculation of this value. METHODS: The volume of left kidney, geometrically more regular, and spleen, with its peculiar shape, of ten animals was established in cubic centimeters after complete immersion in water with the aid of a beaker graduated in millimeters. These values were compared with those obtained by calculating the same volume with a specific mathematical formula: V = 4/3 × π × (r1 x r2 x r3). Data were compared and statistically analyzed by Student's t test. RESULTS: Although the average volume obtained was higher through the direct method (1.13 cm3 for the left kidney and 0.71 cm3 for the spleen) than the values calculated with the mathematical formula (0.81 cm3 and 0.54 cm3), there were no statistically significant differences between the values found for the two organs (p>0.05). CONCLUSION: There was adequate correlation between the direct calculation of the volume of the kidney and spleen with the result of mathematical calculation of these values in the animals' studies.

RACIONAL: O aumento da pressão no sistema venoso portal, induzido pela redução do aporte sanguíneo ao fígado, provoca o desenvolvimento de circulação colateral para que o sangue dos vasos esplâncnicos alcance o sistema venoso da veia cava superior. O baço funciona como um sistema de capacitância que auxilia a compensação do aumento da pressão portal, podendo ocorrer esplenomegalia. OBJETIVO: Reproduzir modelo experimental de hipertensão portal pré-hepática por meio da redução do calibre da veia porta do rato, permitindo o estudo das alterações de volume do baço nesta situação. MÉTODO: Foram utilizados 15 ratos da linhagem Wistar, divididos em três grupos de cinco. Todos os animais foram operados; os primeiros cinco foram utilizados como controle (grupo 1), e reoperados após 15 dias (sham-operated), assim como cinco animais onde foi induzida a hipertensão portal (grupo 2). Os cinco últimos animais, também com hipertensão portal, foram reoperados após 30 dias (grupo 3). A hipertensão portal do tipo pré-hepática foi provocada através da ligadura parcial da veia porta no hilo hepático. O baço foi medido durante a primeira e a segunda operação, permitindo o cálculo de seu volume nos três grupos estudados através de fórmula matemática específica. RESULTADOS: Não foram encontradas diferenças macroscópicas significativas ou alterações maiores do volume do baço quando foram comparados os animais que foram reoperados após 15 dias (grupo 2) e os reoperados após 30 dias (grupo 3), sendo considerado o prazo menor (15 dias), suficiente para avaliação das repercussões no volume após a ligadura parcial da veia porta. Embora o volume dos baços estudados tanto no grupo 2 quanto no grupo 3 tenha aumentado após a primeira operação - variando em média de 0,5417 cm³ para 0,6345 cm³ na reoperação, com aumento de aproximadamente 17% entre a primeira e a segunda fase do estudo -, estes resultados não apresentaram significância estatística (p>0,05). CONCLUSÃO: Não houve aumento com significância estatística do volume do baço após a ligadura parcial da veia porta quando se comparou o volume calculado na primeira operação com o volume encontrado na reoperação. Também não houve diferença quando foram comparados os volumes dos baços dos animais do grupo de reoperados após 15 dias, com o grupo reoperado após 30 dias.

BACKGROUND: Pressure increase in the portal venous system, caused by reduction of blood supply to the liver, develops collateral circulation of splanchnic vessel that reaches the venous system of superior vena cava. The spleen works as an auxiliary capacitance system which compensates the increase in portal pressure that may cause splenomegaly. AIM: To reproduce experimental model of pre-hepatic portal hypertension by reducing the caliber of the vein, allowing the study of alterations in the rat's spleen in this situation. METHODS: Fifteen Wistar rats were divided into three groups of five. All animals were operated, being the first five used as control (group 1) and reoperated after 15 days (sham-operated), along with five other animals had portal hypertension induced (group 2). The last five animals, also with portal hypertension were reoperated after 30 days (group 3). Prehepatic portal hypertension was caused in experimental animals by partial ligation of portal vein in hepatic hilum. The spleen was measured during the first and second operations, allowing the calculation of its volume in the three groups studied using specific mathematical formula. RESULTS: No macroscopic changes in spleen were significant when compared the animals reoperated after 15 days (group 2) to the ones after 30 days (group 3), being 15 days considered enough for the evaluation of effects on spleen after partial ligation. Although the volume of spleens studied in groups 2 and 3 have increased after the first operation - ranging on average from 0.5417 cm³ to 0.6345 cm³ at reoperation with increase of approximately 17% in volume between the first and second phase - the rsults were not statistically significant (p> 0.05). CONCLUSION: There was no statistically significant increase in the spleen volume after partial ligation of the portal vein when comparing the calculated volume in the first operation to the volume found at reoperation. There was no difference when comparing the spleen volumes of the animals reoperated after 15 days and the ones reoperated after 30 days.
